SMAC/DIABLO (Second Mitochondria-derived Activator of Caspase/Direct Inhibitor of Apoptosis-Binding
SMAC/DIABLO is a pro apoptogenic mitochondrial protein released to the cytosol in response to various apoptotic stimuli. Released SMAC then interacts and bypasses inhibitors of apoptosis proteins (IAPs), thus allowing the activation of caspases and apoptosis. Furthermore, SMAC peptides also sensitize tumor cells for apoptosis induced by death-receptor ligation or cytotoxic drugs.
In our project, we specifically used SMAC-N7 (the seven N-terminal amino acids AVPIAQK of the mitochondrial protein SMAC) to enhance the antitumor activity of TRAIL(Tumor necrosis factor-Related Apoptosis-Inducing Ligand).
